Can the trademark symbol match the color of your logo or does it need to be black?

The purpose of either trademark symbol (either ® or ™) is to notify your competitor of your claim of trademark. Any choice of color should not interfere with this purpose.

Can I defend a Supplemental Register Trademark?

Yes. Why? The short answer is this: if you want the trademark, you need to create “acquired distinctiveness” by being the only person using the trademark with your goods or services.
